Best Practices: Proper Storage and Reconstitution of Lyophilized Peptides

Maintaining the structural integrity of peptides is essential in scientific research. Peptides are highly sensitive and can be denatured by temperature changes, UV exposure, or improper handling, which renders them inactive. Premier Aminos synthesizes and lyophilizes peptides to the highest purity standards. Once delivered, it is your responsibility to follow proper handling protocols to preserve this purity.


This guide provides best practices for cold chain management, lyophilized peptide storage, and reconstitution to prevent degradation.


Phase 1: Storage of Lyophilized (Dry) Peptides

Peptides are supplied in a lyophilized (freeze-dried) state, which removes water and temporarily halts degradation, allowing for safe transport. Long-term stability, however, depends on precise temperature control.

  • Short-Term Storage (Up to 4 Weeks): Store lyophilized vials in a standard refrigerator (2°C to 8°C) if you plan to use them within a few weeks.

  • Long-Term Storage (Months to Years): For extended storage, keep lyophilized peptides in a freezer. Use -20°C for several months of stability or -80°C for preservation over years.

  • Moisture and Light Control: Store vials in a dark environment to prevent UV degradation. Keep vials tightly sealed. When freezing, use a sealed container with desiccant packets to prevent moisture buildup.


Important: Avoid repeated freeze-thaw cycles. Remove vials from the freezer only when ready to reconstitute.


Phase 2: The Reconstitution Protocol

Reconstitution returns lyophilized powder to a liquid solution. This step is critical, as small errors can damage peptide chains and compromise your sample.


1. Temperature Equilibration

Do not open a cold vial. After removing a vial from cold storage, let it reach room temperature for 15 to 30 minutes before opening. Opening a cold vial causes condensation, which can compromise peptide purity.


2. Choosing the Right Solvent

Select the solvent based on the peptide’s polarity and intended application:

  • Bacteriostatic Water (BAC Water): BAC water, containing 0.9% benzyl alcohol, is preferred for most research applications. It inhibits bacterial growth, allowing multi-use over several weeks without contamination.

  • Sterile Water: Use when absolute purity is required and benzyl alcohol is not acceptable. Without preservatives, peptides must be used immediately or discarded within a few days to avoid contamination.

  • Dilute Acetic Acid / Ammonium Hydroxide: Basic peptides may need a mildly acidic solvent, such as 1% acetic acid, while acidic peptides may require a basic solvent. Check the peptide’s solubility profile if it does not dissolve in BAC water.


3. The Injection Technique

When adding solvent, do not spray it directly onto the lyophilized powder. Angle the syringe so the needle touches the inner vial wall, and slowly depress the plunger to let the solvent flow gently down the glass.


4. Dissolution: DO NOT SHAKE

After adding the solvent, the peptide will begin to dissolve.

  • Do: Gently swirl the vial in a slow, circular motion.

  • Do Not: Do not shake or invert the vial. Vigorous agitation can break peptide bonds and render the compound inactive. If the solution remains cloudy, refrigerate for 15 to 30 minutes to allow natural dissolution.


Phase 3: Post-Reconstitution Storage

Once reconstituted, peptides degrade more rapidly.

  • Continuous Refrigeration: Store reconstituted peptides at 2°C to 8°C. Do not freeze, as ice crystals can damage peptide chains.

  • Shelf Life: With Bacteriostatic Water and proper refrigeration, most peptides remain stable for 14 to 28 days. After this period, degradation may affect experimental results.

  • Sanitization: Wipe the vial’s rubber stopper with a 70% isopropyl alcohol swab before and after each use to maintain sterility.
